Rédigée à l'encre brune sur un feuillet de papier vélin quadrillé. Dans ce document de premier plan pour l’histoire de l’agronomie, le célèbre botaniste et chef de la maison Vilmorin-Andrieux décrit avec une précision scientifique le processus de fécondation du blé et la méthode technique pour réaliser des croisements (hybridation). Henry de Vilmorin est mondialement reconnu pour avoir révolutionné l'agriculture française par la création de variétés de blé plus productives. Ce texte constitue un témoignage direct de ses travaux de sélectionneur. « La fleur du blé consiste essentiellement en trois étamines, longs sacs à double poche renfermant le pollen ou poussière fécondante et en un ovaire surmonté de deux styles plumeux. Ces organes sont renfermés dans la cavité d'un petit berceau, ressemblant un peu à une coquille de moule et formée d'une coquille creuse et d'une plate, qui sont les balles du blé. La fécondation s'opère naturellement dans le blé par le renversement des étamines, qui répandent alors sur les pistils la poussière dont elles sont remplies. Quand on veut faire un croisement il faut entrouvrir une fleur près de se féconder, supprimer les trois étamines encore vertes et closes, puis refermer la fleur. Le lendemain ou quelques heures après on revient avec un épi du blé qui doit fournir le pollen et choisissant des étamines qui s'apprêtent à répandre leur pollen, on en verse le contenu sur les pistils à féconder. L'opération demande une certaine légèreté de main, mais elle ne présente pas de difficulté réelle. » Ce manuscrit est un témoignage direct des travaux de Henry de Vilmorin (1843-1899), figure centrale de la maison Vilmorin-Andrieux et pionnier de la génétique appliquée. Il a été l'un des premiers à appliquer des méthodes scientifiques rigoureuses pour la création de nouvelles variétés de blé par croisement (notamment le célèbre blé 'Dando'). Le texte décrit avec une précision pédagogique la technique de l'émasculation et de la pollinisation manuelle, qui sont les bases de l'hybridation moderne.
